A Study on the Proliferative Activity of Normal Granulocyte-Monocyte Precursors Using Serial Bone Marrow Culture

Abstract
The proliferative activity of granulocyte-monocyte precursors can be studied in vitro
by semisolid agar culture technique. In the presence of colony stimulating factor, these
cells (CFU-GM) proliferate and produce colonies and clusters of granulocytes and
monocytes. By the way it is difficult to find out reports on the culture of normal
CFU-GM in a strict sense. The present study included ten cases of normal volunteers
and normal donors for bone marrow transplantation which were cultured serially for 28
days. The results are as follows :
1) At 7 day of culture, the mean number of colony and cluster 229 and 481/2¡¿
105 mononuclear cells respectively. So the cluster/colony ratio was 2.18
and the microcluster/macrocluster ratio was 2.28.
2) Colonies were slightly increased in the number at 14 day and markedly decreased
at 21,28 day. But decrease of clusters continued since 7 day of culture.
3) At 7 day of culture, the majority of colony cells are granulocytic forms in
morphology, although monocytic colonies could not be differentiated in immature forms
with acetoorcein in Situ Stain.
4) At 14 day of culture, granulocytic colonies were composed of band or
polymorphonuclear forms. Monocytoid colonies were proportionally increased. The size of
colonies was increased and marginal colony cells had a tendency to be dispersed
outwards.
5) At 21 day of culture, the majority of granulocytic colony cells were degenerated
and monocytoid colony cells showed the morphology of macrophages. At 28 day of
culture, multinucleate macrophages were occasionally observed in remaining colonies or
clusters.
